Every cell division, however, leaves a record: somatic mutations acquired in the earliest blastomeres are inherited by all of their descendants and persist throughout life. Post-mortem tissues collected across the body therefore preserve a retrospective record of early human development. Using post-mortem tissues sampled across the body from multiple individuals, we combined clonal expansion of single cells with deep, multi-site targeted sequencing to reconstruct early lineage trees at high resolution. In this talk, I will discuss how early blastomeres contribute unequally to different organs, how their descendants are distributed across the body, and what these patterns reveal about the organization of early human development. I will also address the implications of these findings for interpreting mosaic disorders and for designing tissue sampling in clinical diagnosis.
