More recent generations of CRISPR variants offer invaluable properties for robust and multiplexed regulation of gene expression. Here the development of Cas12 and Cas13 derived CRISPR tools are discussed alongside a computational tool for facilitating the targeting crRNA design and assembly. We are subsequently employing Cas12 to systematically screen the influence of small nucleolar RNAs, responsible for laying modifications on ribosomal RNA, to assess for impacts on cell fate. Extending this exploration of epitranscriptomic influences on translational regulation, I will touch on a method I have been developing looking to directly pair epitranscriptomic information between ribosomal RNA and mRNA to assess how the two interact with one another.
