To this end, we build on the iCLIP technology to map protein-RNA interaction sites throughout the transcriptome. We have set up ´in vitro iCLIP´ that combines the strength of biochemical experiments with the power of high-throughput sequencing, and we developed a massively parallel reporter screen to identify all cis-regulatory elements that control selected splicing decisions. This experimental setup enables us to obtain mechanistic insights from a global perspective to dissect the regulation and evolution of critical splicing events. We also use our approaches to understand the impact of human genome variation on gene regulation and to predict mutations that cause mis-splicing in cancer and neurodegenerative diseases.
