Piwi-interacting RNAs (piRNAs) engage Piwi proteins to suppress transposons and non-self nucleic acids and maintain genome integrity and are essential for fertility in a variety of organisms. My Phd thesis is about biogenesis of piRNAs in C. elegans. I will present two complexes engaged in piRNA biogenesis. One is USTC, upstream sequence transcription complex which mediates the transcription of piRNA precursors. The other one is PICS (piRNA biogenesis and chromosome segregation) complex that locates in the peri-nuclear granules and promotes piRNA maturation in cytoplasm and is also required for faithful chromosomal segregation.
