However, evidence suggests that many cytoplasmic, nuclear, and membrane proteins can also reach the cell surface via Golgi-bypassing routes, collectively referred to as unconventional protein secretion (UPS). Several transmembrane proteins, including CFTR, CD45, Kv4, Mpl, pendrin, and SARS-CoV-2 Spike, are known to reach the cell surface via UPS under specific conditions. Understanding UPS pathways is important not only for elucidating intracellular trafficking mechanisms but also because many proteins involved in UPS are associated with human diseases. However, the molecular mechanisms underlying UPS remain largely elusive. The ER is highly dynamic in terms of its synthesis, degradation, and shape. One of the processes that leads to dynamic change in ER structures is ER-phagy, which is facilitated by ER-phagy receptors. These ER-phagy receptors have the ability to induce changes in ER morphology by sensing and forming ER membrane curvature. In this talk, I will introduce the molecular machinery involved in the UPS of transmembrane proteins, particularly those related to ER-phagy receptor proteins, and discuss the potential for new therapeutic strategies to treat UPS-associated diseases.
